--Genetic transformation of walnut is accomplished by cultivating engineered Agrobacterium tumefaciens with repetitively embryogenic somatic embryos derived from anther tissue or immature zygotic embryos. Transgenic embryos are selected through several generations on kanamycin medium; gus activity is the preliminary indicator of transformation. In addition to these markers we have also inserted genes for insect resistance (Bt), ftmgal resistance (chitin binding), rootability (rolABC) and others. Initial efficacy tests are based on embryo response to the appropriate insect or fungus. Potent lines are then micropropagated and budded on to rootstock for field trials (several are underway). Major hindrances include genetic stability of the embryo clone in culture and various aspects of propagation.
