Labelled synthetic lignin 14 C-ring-labelled DHP) was chemically coupled to the lignin in kraft pulp using potassium ferricyanide at pH 10. This 14 C-labelled pulp was used in a quick and sensitive method for evaluating ligninolytic enzymes and mediators. The labelled pulp has a high specific radioactivity, that gives reliable measurements of cpm in buffers and in NaOH-extracts, when 30 mg of pulp is incubated with enzyme in 1.5 ml buffer in an Eppendorf tube. In a screening program a great number of enzymes and the mediators HBT and ABTS were tested. Also pH-optima against 2,6-DMP (2,6-dimethoxyphenol), ABTS [2,2’-azinobis-( 3-ethylbenzthiazoline-6-sulphonate)] and HBT (l-hydroxybenzotriazole) with and without pulp are reported.
