My TAPPI Story: Naomi Gehling
The activities of peroxidase, glucose oxidase and catalase were measured in cultures of Trametes versicofor and Pleurotus eryngii in trophophase and during the onset of idiophase. Both fungi were grown on the Fahraeus-Reinhammar medium with glucose as the main source of carbon and asparagine as the main source of nitrogen. Transition to idiophase was provoked by means of exchange of the medium for one depleted of glucose or asparagine. It was observed that in the Trametes versicolor cultures depleted of the carbon source activity of the peroxidase rose both in mycelia and the culture filtrate in comparison to the trophophasic cultures, whereas the activity of catalase was high in trophophasic cultures and dropped in these depleted of the carbon source. In the Pleurotus eryngii cultures activities of peroxidase and glucose oxidase changed similarly to these in Trametes cultures while catalase activity was lacking in the culture filtrate in all cultures analysed. We concluded that the correlated changes of peroxidasic and glucose oxidase activities are a part of pleiotropic effect resulting from the onset of idiophase whereas inversely oriented changes of catalase activity resulted from an adaptation of the fungi to ligninolysis by peroxidases demanding all of the hydrogen peroxide available in the cultures.
