Efficient production of transgenic Eucalyptus camnldu/ensis has been achieved through Agrobncterium-mediated transformation. A simple and effective organogenesis system was developed using a modified B5C medium for both callus induction from hypocotyl explants and shoot regeneration from the calli. Rooting ability of transformed shoots obtained from antibiotics selection was markedly improved with the employment of a pre-rooting culture on medium containing a low level of cytokinin. Transgenic plants harboring GUS reporter gene were generated and confirmed for T-DNA integration and reporter gene expression. This transformation and regeneration system has been modified further for field-grown elite eucalyptus trees. Further studies are currently underway for transformation with lignin biosynthetic pathway genes in order to produce transgenic Eucn!vplu.s with modified lignin properties.
