Detection of Lignolytic Enzymes Produced by Phlebia subserialis and a Comparative Analysis on Picea abies with Other White-Rot Fungi, 2003 Fall Technical Conference

White-rot fungi, with their unique ability to completely mineralize recalcitrant nonphenolic lignin moieties, hold promise in the biotechnology field. The main objective of this study was to extend research on Phlebia subserialis as a biopulping fungus by documenting the lignolytic enzymes produced and making physicochemical comparisons with both C. subvermispora and T. versicolor. This is the first report on the constitutive and induced production of extracellular lignolytic enzymes of P. subserialis. In this study, P. subserialis produced 4.47 nkatal ml-1 laccase activity measured by the oxidation of ABTS and 0.240 nkatal ml-1 manganese peroxidase measured by the oxidation of DMP, induced by xylidine and under optimized laboratory conditions using a semi-defined medium. Both T. versicolor and C. subvermispora produced typical enzyme activity under the same induced conditions. P. subserialis constitutively produced both laccase and manganese peroxidase in a novel experiment extracting enzymes from chips, under biopulping conditions. The results of this work indicate that the organisms studied are selective lignin degraders based on Klason lignin and NMR carbohydrate chemical analysis during the 4-week treatment period.
